1. Filling Column Chromatography
Filling columns can use any type of tubing as long as it is: clean, inert to the sample, and able to withstand the oven temperature of the GC
2. Classification
Classification by stationary phase polarity:
1). Non-polar: 100% dimethyl polysiloxane.
2). Weak polarity: 5% phenyl-methyl polysiloxane.
3). Moderate polarity: 35% phenyl-methyl polysiloxane, etc.
4). Strong polarity: polyethylene glycol, polyethylene glycol and nitrodiethylene terephthalate, etc.
3. Selection of chromatographic column
1). The choice of the polarity of the fixative (according to the principle of similar compatibility)
A. Non-polar fixative → dissolve in order of boiling point
B. Polar fixative → When the boiling point is the same, dissolve in the order of polarity from small to large
2). Selection of the concentration of the fixative and the film thickness of the capillary column
A. For low boiling point compounds
High concentration (10%30%)
High film thickness (1~5um)
B. For high boilers
Low concentration (1%~5%)
Low film thickness (0.25~0.5um)
4. Column Affecting Factors
1). Influence of inner diameter: the smaller the inner diameter, the better the degree of separation.
2). Influence of film thickness: the smaller the inner diameter, the better the separation.
5. Chromatographic Column – column temperature
The stationary phase coating inside the column has an applicable temperature range.
6. Column Aging
1). The new column contains solvents and high-boiling substances, so the baseline is unstable, ghost peaks and noise appear; the same problem exists when the old column has not been used for a long time.
2). Generally, heating aging is adopted, that is, the temperature is programmed from room temperature to the highest temperature, and kept in the high temperature section for several hours. When the new column is aging, it is best not to connect the detector.
